BPC-157 is a synthetic 15-amino-acid peptide — a partial sequence of Body Protection Compound, a protein isolated from human gastric juice. The published literature reports a defined cascade: VEGFR2 receptor activation, then Src/FAK and Akt, then eNOS and nitric-oxide bioavailability — alongside a fibroblast branch (FAK–paxillin signaling, F-actin formation) and an early transcriptional burst, with egr-1 expressed by 15 minutes and its own repressor nab2 arriving almost immediately after. The reviewing group proposes that self-limiting loop as the reason repair is rapid but not fibrotic.
The peptide's signature is balance, not one-way stimulation: in rat models it counteracts a blocked NO system (L-NAME) and an over-stimulated one (L-arginine) alike, with free-radical formation falling alongside — and angiogenesis is promoted in tendon, ligament and muscle models while opposed in the cornea, where transparency depends on keeping vessels out.
Acemannan approaches repair from the other direction. An acetylated polymannose from the inner gel of Aloe vera, it is recognized by pattern-recognition receptors of the innate immune system — the mannose receptor, TLR-4 and TLR-5 — and in gingival-fibroblast studies it raised proliferation and upregulated KGF-1, VEGF and type I collagen.
